just eyeball it. wrap them on as straight as you can, then look down the rod and tweak them where necessary. you can pull a thread from top to bottom for the initial line if you want, but its not needed.
sounds like youre getting closer. dont forget to take a picture or two as its coming together and post them when youre done!

also if you can, mount the reel you plan to use on this rod in the reel seat to align the stripper guide. yould be surprised how different reels mount at slightly different angles in the same seat
